RESUMO

The real-time reverse transcription-polymerase chain reaction (real-time RT-qPCR) has become a leading technique for the detection and quantification of arboviruses, including Chikungunya, Dengue, and Zika viruses. In this study, an updated real-time RT-qPCR assay was designed and evaluated together with a synthetic positive-control chimeric RNA for the simultaneous detection and quantification of Chikungunya, Dengue, and Zika viruses. Amplification assays were performed to verify the construct integrity and optimal reaction/thermal cycling conditions. The analytical sensitivity of the assay was determined for each virus in single and multiplex reactions, as well as the performance in the detection and viral load quantification of experimental samples. The real-time RT-qPCR assay presented here allowed for the simultaneous detection and quantification of Chikungunya, Dengue, and Zika viruses and could be applied in several studies where the accurate quantification of viral genomes is required.

Resumen A pesar de algunas limitaciones éticas, los animales juegan un papel importante como anfitriones sustitutos para investigar los mecanismos fisiopatológicos de enfermedades con el fin de indagar en ellos medicamentos contra diferentes patologías. Uno de los grandes problemas en salud pública a nivel mundial en el contexto farmacológico es la producción de antibióticos y la ocurrencia de resistencia microbiana, además, cada vez resulta más complejo el uso de modelos animales por las restricciones bioéticas actuales, no obstante, es necesario usar modelos simples en los estudios preliminares que permitan evaluar las interacciones huésped-patógeno-antimicrobiano. Al validar que Caenorhabditis elegans es susceptible a varias bacterias y además tiene la capacidad de responder a estímulos ambientales con cambios observables en el comportamiento tras ser alimentado con diversas bacterias, resulta muy útil usarlo en este tipo de investigaciones ya que tiene una vida promedio corta y no cuenta con restricciones éticas para su uso. Por lo anterior, en este artículo se revisa la susceptibilidad que tiene C.elegans de infectarse con diferentes bacterias, además, ya que aún no se ha validado completamente como modelo para poner a prueba antimicrobianos se propone que este nematodo es útil como modelo In vivo para evaluar infecciones y tratamientos antibacterianos.


Abstract Despite some ethical limitations, animals play an important role as surrogate hosts in investigating the pathophysiological mechanisms of disease in order to test drugs against different pathologies. One of the great problems in public health worldwide in the pharmacological context is the production of antibiotics and the occurrence of microbial resistance, the use of animal models is becoming increasingly complex due to current bioethical restrictions, however, it is necessary to use models simple in preliminary studies that allow evaluating host-pathogen-antimicrobial interactions. Validating that Caenorhabditis elegans is susceptible to various bacteria and also has the ability to respond to environmental stimuli with observable changes in behavior after being fed with various bacteria, it is very useful to use it in this type of research since it has a short average life and does not have ethical restrictions for its use. Therefore, this article reviews the susceptibility of C. elegance to become infected with different bacteria, in addition, since it has not yet been fully validated as a model to test antimicrobials, it is proposed that this nematode is useful as an in vivo model. to evaluate infections and antibacterial treatments.

Resumen Los canales de calcio son proteínas de membrana que constituyen la vía más importante para el ingreso del ion calcio (Ca2+) a la célula. Al abrirse, permiten el ingreso selectivo del ion, iniciando una variedad de procesos como contracción muscular, secreción endocrina y liberación de neurotransmisores, entre otros. Estas proteínas se agrupan en tres categorías de acuerdo con sus propiedades estructurales y funcionales: (i) Canales de Ca2+ operados por interacción receptor-ligando (ROCC), (ii) Canales activados por parámetros físicos (Transient Receptor Potencial, TRP) y (iii) Canales de Calcio dependientes de voltaje (VDCCs), siendo estos últimos los más estudiados debido a su presencia en células excitables. Dada la importancia de Ca2+ en la fisiología celular, los canales de Ca2+ constituyen un punto de acción farmacológica importante para múltiples tratamientos y, por tanto, son objeto de estudio para el desarrollo de nuevos fármacos. El objetivo de esta revisión es explicar la importancia de los canales de Ca2+ desde una proyección farmacológica, a partir de la exploración documental de artículos publicados hasta la fecha teniendo en cuenta temas relacionados con la estructura de los canales Ca2+, sus propiedades biofísicas, localización celular, funcionamiento y su interacción farmacológica.


Abstract Calcium channels are membrane proteins that constitute the most important route for the entry of the calcium ion (Ca2+) into the cell. When opened, they allow selective ion entrance, starting a variety of processes such as muscular contraction, endocrine secretion and neurotransmitters release, among others. These proteins are classified in three categories according to their structural and functional properties: (i) Receptor-operated calcium channels (ROCC), (ii) Channels activated by physical parameters (Transient Receptor Potential or TRP-channels) and (iii) Voltage-dependent calcium channels (VDCCs), the latter being the most studied due to its presence in excitable cells. Given the importance of Ca2+ in the cellular physiology, the calcium channels constitute targets for pharmacological action for multiple treatments, and therefore, they are object of study for the development of new medicaments. The objective of this review is to explain the importance of the channels of Ca2+ from a pharmacological projection, by exploring the articles published, bearing in mind topics related to the structure of the channels Ca2+, properties of their biophysics, cellular location, functioning and their pharmacological interaction.

Resumen La nueva pandemia por SARS-CoV-2 ha incentivado la búsqueda de alternativas terapéuticas que reduzcan los índices de mortalidad. Dentro de estas, el uso de péptidos se destaca por su facilidad de síntesis, fácil producción a gran escala, especificidad en la respuesta inmunológica, versatilidad en la dosificación, alta pureza, economía, entre otros. Esto ofrece una excelente opción al enfoque tradicional, con lo que se resta la evaluación de su eficacia, eficiencia y seguridad a nivel clínico.


Abstract The new pandemic by SARS-CoV-2 has encouraged the search for therapeutic alternatives that reduce mortality rates; within these, the use of peptides stands out for its ease of synthesis; easy production on a large scale, specificity in the immune response, versatility in dosage, high purity, economy, among others; offering an excellent option to the traditional approach, failing to evaluate its effectiveness, efficiency and safety at clinical level.
